“S.PneumoStrip” test permitting the detection and identification of 42 S. pneumonaie serotypes in the DNA samples extracted from bacterial cultures

Description

S. pneumonaie infections pose a serious public health problem both in industrialised countries and in those which are less developed. It is the cause of high morbidity and mortality, being one of the main agents responsible for a wide variety of clinical conditions, from benign infections ranging from otitis media and acute sinusitis to severe infections including septicaemia, meningitis and pneumonia. Among the most interesting structures of this bacteria is a complex polysaccharide capsule external to the cell wall.  This capsule protects the bacteria from phagocytosis and is indispensable for its virulence.  In fact, the non-capsulated variants of the bacteria are, normally, non-virulent.  The antigenic composition of the capsule varies between the different strains and makes it possible to group S. Pneumonaie into more than 100 different capsular serotypes and approximately 45 serogroups.  Serotypes of crossed immunogenicity are defined as belonging to a same serogroup.  Each serotype is identified by means of an antigen-antibody reaction using specific antiserums, resulting in the swelling of a capsule, a phenomenon known as “Quellung”, a term precisely meaning ‘swelling’ in German.  While this remains the benchmark model, it is a long, complex and laborious technique requiring highly skilled staff to carry it out and its use is therefore restricted to qualified laboratories.  The new S. PneumoStrip is the only test that fast, easily and simply individually identifies 42 different types of S. pneumonaie in DNA samples, extracted from bacterial cultures.  Among the serotypes detected are the 23 included in the different available vaccines.  The 42 serotypes detected are:  1, 2, 3, 4, 5, 6A, 6B, 6C, 6D, 7F/7A, 8, 9N/9L, 9A/9V, 10A, 10B, 10F/C, 11A/D, 11B, 11C, 11F, 12F, 14, 15B/15C, 17F, 18A, 18B/18C, 18F, 19A, 19F, 20, 22F/22A, 23F, 33F/33A.

Applications
